Staining Techniques use in microbiology

Objectives of staining techniques use in microbiology are to examine the morphology, to classify and identify and to examine the different structure.

  • For preparation of bacterial culture smear place a drop of water on a clean glass slide.
  • Take loopful culture.
  • Make a smear.
  • Allow the smear air dry.
  • Fix the smear by passing on the flame.
  • Types of staining methods are Simple staining methods, Differential staining methods and Special staining method.
  • In simple staining method one dye is used and it is used to study the morphology as well as presence or absence of bacteria in smear.
  • In differential staining method more than one dye is used to differentiate.
  • Special staining method is used to demonstrate the presence of certain bacterial structure like spores, capsules, flagella and metachromatic granules.
